      通过测定12种黄酮化合物的抗菌活性,并筛选出与头孢他啶联用时具有协同作用的增敏剂。以微量稀释法测定化合物的最低抑菌浓度,棋盘法测定化合物联用后的最低抑菌浓度。以杀菌曲线法、纸片法、细菌泳动能力法观察化合物联用后对细菌的影响。结果表明,12种化合物均表现出一定的抗菌活性,对金黄色葡萄球菌、枯草芽孢杆菌、铜绿假单胞杆菌、大肠杆菌抑菌率均低于30%。单一黄酮化合物与头孢他啶联用时,联合抑菌率有不同程度的提高,但未达到协同作用。而当盐酸黄酮哌酯、3-甲基黄酮-8-羧酸两者组合后与头孢他啶联用时,可以显著增强头孢他啶抗金黄色葡萄球菌的能力,三者具有协同作用,联合抑菌率为95%。同时,盐酸黄酮哌酯、3-甲基黄酮-8-羧酸两者组合可以增强头孢他啶抗枯草芽孢杆菌、铜绿假单胞杆菌、大肠杆菌、耐甲氧西林金黄色葡萄球菌的作用,联合抑菌率分别为81%、78%、78%、82%,所以未达到协同作用。盐酸黄酮哌酯、3-甲基黄酮-8-羧酸两者同时与头孢他啶联用时可以增强头孢他啶抗金黄色葡萄球菌的能力,具有协同作用。

      By measuring the antibacterial activity of 12 flavonoids and screening out sensitizers in combination with ceftazidime. The minimum inhibitory concentration of the compound was determined by microdilution method. Chessboard method was used to determine the minimum inhibitory concentration after combined use of compounds. The influence of compound combination on bacteria was observed by time-kill curve, disk diffusion, swimming motility. The results showed that 12 flavonoids had certain antibacterial activity, and the inhibition rates against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa and Escherichia coli were less than 30%. When a single flavonoid compound was combined with ceftazidime, its inhibitory rate increased to varying degrees, but did not achieve a synergistic effect. The combination of flavonidate hydrochloride and 3-methylflavone-8-carboxylic acid combined with ceftazidime could significantly enhance the ability of ceftazidime to anti-staphylococcus aureus, and the three had synergistic effect. The combined bacteriostatic rate was 95%. The combination of flavonidate hydrochloride and 3-methylflavon-8-carboxylic acid can enhance the anti-bacterial rate of ceftazidime against Bacillus subtilis, Pseudomonas aeruginosa, Escherichia coli and methicillin-resistant Staphylococcus aureus ,they were 81%、78%、78% and 82%, respectively, so they did not achieve synergy. Both flavone hydrochloride and 3-methylflavon-8-carboxylic acid can enhance the ability of ceftazidime against Staphylococcus aureus when used in combination with ceftazidime, and have a synergistic effect.
